A while back there used to be a number of commercial applications designed to enable you to paint more freely, like a traditional artist, rather than ‘by pixels’. Image these days is based around editing and enhancing photos, so the latest commercial apps are based around this, rather than creating your own image, from scratch.

TwistedBrush is an application designed for those who require more freedom to create artistic works using their computer from absolutely nothing. The application supports drawing tablets and over 5,000 different brush styles as well as a number of popular features such as, tracing, masks, filters, guides and many more. With TwistedBrush, you can quickly design your own cartoons, manga images and more. You can also touch-up your own or other images using this tool.

Note that this is the free version. A commercial version is available with additional functionality, more brushes and a useful brush editing tool.
